Synthesis, Characterizations, Anticancer Activities and In-silico studies of Pyrazolealdehydes Based Curcumin-I Derivatives

Abstract: Vilsmeier–Haack reaction was used to prepare different substituted pyrazolealdehydes, which were attached to the curcumin-I via Knoevenagel condensation. Further, the disemicarbazone derivatives of the condensates were prepared. All these compounds were characterized and purified by different spectroscopic and chromatographic techniques. The anticancer activities of the compounds on MCF-7 cell line were in the range of 59-79% (at 1.0 µg/mL). In-silico studies indicated that pyrazolealdehydes preferred to enter minor groove, whereas Knoevenagel condensates and their corresponding disemicarbazones favored major grooves of DNA.
